dc.description.abstract | La industria alimentaria debe asegurar a las personas consumidoras que el producto que se les ofrece no sufre adulteraciones de ningún tipo. Ello se garantiza mediante diferentes normativas tanto en el marco legislativo europeo como en el estatal. En el caso de los productos cárnicos procesados, el fraude se produce, en la mayoría de los casos, por sustitución de especies. Para poder controlar este problema, existen diferentes técnicas analíticas que permiten identificar especies animales presentes en los alimentos. Así, el objetivo del presente trabajo ha sido poner a punto una metodología para la identificación de las especies vacuno y equino basada en la PCR a tiempo real que sea rápida, sencilla y de elevada especificidad. En primer lugar, se determinó la técnica más adecuada para la obtención del material genético, resultando seleccionado como óptimo, de entre los procedimientos ensayados, el kit comercial NucleoSpin FOOD-colum. Para la detección de ADN de las especies en estudio, se seleccionaron y diseñaron cebadores que amplifican una secuencia de los genes Cyclic-GMP-phospodiester y Growth hormone receptor (GHR) para vacuno y equino respectivamente. Para poner a punto la técnica de PCR a tiempo real, se realizaron diferentes ensayos para optimizar las condiciones de amplificación y calcular las eficiencias de los genes seleccionados. Además, se llevaron a cabo los ensayos necesarios para determinar la sensibilidad y la especificidad de la técnica, dando como resultado un protocolo que puede emplearse para la detección e identificación de ADN de vacuno y equino en productos cárnicos. | es_ES |

Arazo hori kontrolatzeko, hainbat teknika analitiko daude, zeinak elikagaietako animalia-espezieak identifikatzea ahalbidetzen duten. Hala, lan honen helburua da behi- eta zaldi-espezieak identifikatzeko metodologia bat prestatzea, denbora errealeko PCRan oinarritua, azkarra, sinplea eta berezitasun handikoa. Lehenik eta behin, material genetikoa lortzeko teknikarik egokiena zein den zehaztu zen, eta, probatutako prozeduren artean, hoberentzat hautatu zen NukleeoSpin FOOD-colum kit komertziala. dc.description.abstract | The food industry must assure consumers that the product offered to them is not adulterated in any way. This is guaranteed by different regulations in both the European and national legislative frameworks. In the case of processed meat products, fraud is produced, in most cases, by substitution of species. In order to control this problem, there are different analytical techniques that make it possible to identify the animal species present in foods. Thus, the objective of the present work was to develop a methodology for the identification of DNA from bovine and equine species based on real-time PCR, as it is fast, simple and of high specificity. First, the most appropriate technique for obtaining the genetic material was determined, and the commercial NucleoSpin FOOD-colum kit was selected as the best among the procedures tested. For the detection of DNA from the species under study, primers that amplify the sequences of the Cyclic-GMP-phospodiester and Growth hormone receptor (GHR) genes for bovine and horses, respectively, were selected and designed. To ser the appropriate conditions for the real-time PCR technique, different assays were performed to optimize the amplification conditions and to calculate the efficiencies of the selected genes.
