Entre los métodos de control de malas hierbas, los herbicidas aparecen como métodos rápidos y
efectivos, y conforman el método más utilizado para el control de malas hierbas. Dos de los
grupos de herbicidas más importantes son los inhibidores de la 5-enolpiruvilsiquimato-3-fosfatosintasa (EPSPS) y la acetolactato sintasa (ALS). El único inhibidor de la EPSPS que se conoce es el glifosato, el herbicida más utilizado a nivel mundial; mientras, los inhibidores de la ALS incluyen
numerosas sustancias activas. La EPSPS y la ALS son enzimas importantes que forman parte de la
biosíntesis de aminoácidos aromáticos y aminoácidos ramificados, respectivamente. Cómo el
glifosato o los inhibidores de la ALS bloquean estas rutas de biosíntesis de aminoácidos está
estudiado a fondo, pero la secuencia de eventos fisiológicos que ocurre entre la aplicación del
herbicida y la muerte de la planta no se conoce del todo. Los efectos fisiológicos establecidos
hoy en día para ambos grupos de herbicidas son bastante parecidos (a pesar de que las enzimas
diana sean diferentes). Algunos estudios muestran que entre estos efectos fisiológicos
provocados por el glifosato y los inhibidores de la ALS está el estrés oxidativo, pero su origen (si
está ligado a la inhibición de la EPSPS o ALS o es un efecto secundario aparte) y la importancia
del daño oxidativo y las alteraciones en los sistemas antioxidantes (como el glutatión) inducidos
en la cascada fisiológica que termina con la muerte de la planta no se conocen. En este contexto, el objetivo principal de esta tesis fue evaluar el papel del estrés oxidativo y el metabolismo del glutatión en el modo de acción del glifosato y los inhibidores de la ALS,
tratando de elucidar si está relacionado con el mecanismo de acción de ambos grupos de
herbicidas. Se crecieron cuatro poblaciones de la mala hierba Amaranthus palmeri S. Wats.:
sensible a glifosato (GFS), resistente a glifosato (GFR), sensible a inhibidores de la ALS (AIS) y
resistente a inhibidores de la ALS (AIR). En las dos poblaciones resistentes, la resistencia venía
dada por mecanismos target-site (TSR). Las poblaciones GFS y GFR fueron tratadas con
diferentes dosis de glifosato, mientras que las poblaciones AIS y AIR fueron tratadas con dosis
diferentes de nicosulfuron (una sulfonilurea, inhibidor de la ALS). Se realizaron varios análisis
fisiológicos y se compararon, en las hojas de estas poblaciones: acumulación de especies
reactivas de oxígeno (ROS), daño oxidativo, sistemas antioxidantes (con especial énfasis en el
metabolismo del glutatión) y señalización hormonal.Casi no hubo diferencias entre las plantas sensibles y resistentes no tratadas en los parámetros
analizados, mostrando que la TSR no viene acompañada por un estado oxidativo basal diferente.
Las plantas GFS y AIS tratadas murieron con todas las dosis de glifosato o nicosulfuron; en
cambio, todas las plantas resistentes sobrevivieron. Las plantas GFS y AIS tratadas mostraron
una producción y acumulación de ROS mayor que las no tratadas, un efecto que no ocurrió en las
resistentes. Esta sobreproducción de ROS en las plantas sensibles tratadas provocó una
peroxidación lipídica proporcional a la dosis de herbicida, un indicador inequívoco de estrés
oxidativo, y cambios en los sistemas antioxidantes. Solo las plantas sensibles de las dos
poblaciones mostraron un incremento de la actividad y expresión génica de la glutatión Stransferasa (GST), indicando la implicación del glutatión como antioxidante. La presencia de estrés oxidativo solo en las plantas sensibles tratadas con glifosato o
nicosulfuron y su ausencia en las plantas TSR indica que el estrés oxidativo está relacionado con
la inhibición de la EPSPS o la ALS, aunque los mecanismos exactos permanecen sin esclarecer. El
daño oxidativo se probó como otro efecto común de ambos herbicidas, pero fue demasiado leve
para provocar por sí solo la muerte de la planta. Por otra parte, los dos herbicidas alteraron el perfil hormonal, pero indujeron hormonas
diferentes. Así, los cambios observados en el perfil hormonal muestran un efecto diferente de la
acción herbicida. Los mecanismos que provocan estos cambios hormonales siguen siendo
desconocidos, pero la inducción de ciertas hormonas puede estar relacionada con el estrés
oxidativo. [--]

fisiologiko hauen artean dagoela erakusten dute, baina honen jatorria (EPSPS edo ALS
inhibizioari lotuta badagoen ala bigarren mailako efektu bat baden) eta oxidazio-kalteak eta
sistema antioxidatzaileetan (hala nola, glutationean) eragindako alterazioek landarearen
heriotzarekin bukatzen den erorketa fisiologikoan duten garrantzia ez dira ezagutzen.
Testuinguru honetan, honako tesi honen helburu nagusia oxidazio-estresak eta glutationaren
metabolismoak glifosatoaren edo ALS-inhibitzaileen ekintza moduan duten garrantzia
ebaluatzea izan zen, bi herbizida-taldeen ekintza mekanismoarekin erlazionatuta al dagoen
argitzen saiatuz. Amaranthus palmeri S. Wats. belar txarraren lau populazio hazi ziren:
glifosatoarekiko sentikorra (GFS), glifosatoarekiko erresistentea (GFR), ALS-inhibitzaileekiko
sentikorra (AIS), eta ALS-inhibitzaileekiko erresistentea (AIR). Bi populazio erresistenteetan,
erresistentzia target-site (TSR) mekanismo batek eman zuen. GFS eta GFR populazioak glifosato
dosi ezberdinekin tratatu izan ziren; bestetik, AIS eta AIR populazioak nikosulfuroi (sulfonilurea
bat, ALS inhibitzailea) dosi ezberdinekin tratatu izan ziren. Zenbait analisi fisiologiko egin eta
alderatu ziren lau populazio hauen hostoetan: oxigenoko espezie erreaktiboen (ROS)
akumulazioa, oxidazio-kaltea, sistema antioxidatzaileak (enfasi bereziarekin glutationaren
metabolismoan) eta hormona-seinaleztapena. Ia ez zegoen desberdintasunik tratatu gabeko landare sentikorren eta erresistenteen artean
aztertutako parametroetan, TSR-a oinarrizko egoera oxidatibo desberdin batekin batera ez
datorrela frogatuz. Tratatutako GFS eta AIS landareak hil ziren glifosato edo nikosulfuroi dosi
guztiekin; landare erresistente guztiek bizirik iraun zuten, ordea. Tratatutako GFS eta AIS
landareek ROS ekoizpen eta akumulazio handiagoa erakutsi zuten tratatu gabekoek baino,
erresistenteetan ageri ez zen efektu bat. Tratatutako landare sentikorretan azaldutako ROS
gehiegizko ekoizpen handi honek herbizida dosiarekiko lipidoen peroxidazio proportzionala
eragin zuen, oxidazio-estresaren adierazle nabari bat, eta aldaerak sistema antioxidatzaileetan.
Soilik bi populazio sentikorretako landareek glutation S-transferasaren (GST) aktibitatearen eta
gene-adierazpenaren igoera erakutsi zuten, glutationaren inplikazioa antioxidatzaile bezala
adieraziz.
Oxidazio-estresaren presentziak bakarrik glifosatoarekin edo nikosulfuroiarekin tratatutako
landare sentikorretan eta oxidazio-estres ezak TSR landareetan oxidazio-estresa EPSPS edo ALS aren inhibizioarekin erlazionatuta dagoela adierazten dute, nahiz eta mekanismo zehatzak
iheskor geratu. Oxidazio-kaltea bi herbiziden beste efektu komun bat bezala frogatu zen, baina
leunegia izan zen berak bakarrik landarearen heriotza eragiteko, oxidazio-estresa landarearen
heriotzaren kausa nagusia ez dela frogatuz.
Halaber, bi herbizidek hormona-profila eraldatu zuten, baina hormona desberdinak induzitu
zituzten. Horrela, hormona-profilean aurkitutako aldaketek herbizida-akzioaren efektu

Among the available methods to control weeds, herbicides stand as quick and effective tools,
and conform the most extended method to control weeds. Two of the most prominent
herbicide groups are 5-enolpyruvylshikimate-3-phosphate-synthase (EPSPS) and acetolactate
synthase (ALS) inhibitors. The only known EPSPS inhibitor is glyphosate, the most used herbicide
in the world, while ALS inhibitors include many active substances. EPSPS and ALS are important
enzymes involved in the synthesis of aromatic amino acids and branched-chain amino acids,
respectively. How these amino acid biosynthetic pathways are disrupted after the application of
glyphosate or an ALS inhibitor is well studied, but the sequence of physiological events from
herbicide application to plant death is not completely understood. Currently established
physiological effects are quite similar between both herbicide groups (despite the different
target enzymes). Some studies show that oxidative stress is among these physiological effects
induced by glyphosate and ALS inhibitors, but both its origin (whether it is linked to EPSPS or
ALS inhibition or is a separate side effect) and the role of oxidative damage and alterations of
the antioxidant systems (such as glutathione) in the physiological downfall that ends with plant
death are unknown. In this context, the present study mainly aimed to evaluate the role of the oxidative stress and
the metabolism of glutathione in the mode of action of glyphosate or ALS inhibitors, trying to
unravel if they are related to the mechanism of action of both groups of herbicides. Four
populations of the weed Amaranthus palmeri S. Wats. were grown: glyphosate-sensitive (GFS),
glyphosate-resistant (GFR), ALS-inhibitor sensitive (AIS), and ALS-inhibitor resistant (AIR). In
both resistant populations, resistance was conferred by a target-site resistance (TSR)
mechanism. GFS and GFR populations were treated with different glyphosate doses, while AIS
and AIR populations were treated with different nicosulfuron (a sulfonylurea, ALS inhibitor)
doses. Several physiological analyses were performed and compared in leaves of these
populations: reactive oxygen species (ROS) accumulation, oxidative damage, antioxidant
systems (with special emphasis on glutathione metabolism), and hormonal signalling. There were virtually no differences between untreated sensitive and resistant plants in the
measured parameters evidencing that TSR is not accompanied by a different basal oxidative
pattern in any of both cases. GFS and AIS plants treated with any dose of glyphosate or nicosulfuron died, while all resistant plants survived the herbicide treatments. Treated GFS and
AIS plants showed a higher ROS production and accumulation than untreated plants, an effect
that did not take place in resistant plants. This great ROS overproduction in treated sensitive
plants led to a herbicide dose-proportional lipid peroxidation, an unmistakable sign of oxidative
damage and to changes of antioxidant systems. Only treated plants of both sensitive
populations showed increase of glutathione S-transferase (GST) activity and gene expression,
indicating the involvement of glutathione as an antioxidant. The presence of oxidative stress only in sensitive plants treated with glyphosate and an ALS
inhibitor and its absence in TSR plants indicate that oxidative stress is related to EPSPS or ALS
inhibition, although the exact mechanisms remain elusive. Oxidative damage was evidenced as
another common effect of both types of herbicides, but it was too mild to provoke plant death
by itself, evidencing that oxidative stress is not the main cause of plant death. Apart from that, both herbicides affected hormonal profile, but induced different hormones. In
this way, the changes detected in the hormonal profile show a different effect of herbicidal
action. The mechanisms by which these hormonal changes are triggered remain unknown, but
the induction of certain hormones may be related to oxidative stress. [--]
