El control químico de malas hierbas a través de herbicidas ha sido ampliamente utilizado
en la agricultura, pero el uso continuo y excesivo desde mediados del siglo XX ha resultado en
poblaciones de malas hierbas resistentes a los herbicidas. Así pues, es importante conocer el
mecanismo de resistencia de estas malas hierbas resistentes, para poder utilizar métodos de
control efectivos frente a las malas hierbas, y evitar la evolución de otras resistencias.
Amaranthus palmeri es una mala hierba originaria de los EE. UU., y debido a su fisiología
y su capacidad para propagarse e invadir, se considera una planta muy problemática e invasora.
Además, se han encontrado numerosas poblaciones resistentes, como a los herbicidas
inhibidores de la enzima acetolactato sintasa (ALS) en la ruta de biosíntesis de lo aminoácidos
ramificados. Generalmente, el mecanismo de resistencia a estos herbicidas se basa en una diana
menos sensible pero pueden haberse desarrollado otros mecanismos de resistencia no
relacionados con la diana (NTS), como la capacidad de metabolizar el herbicida a través de la
famila de enzimas glutatión-S-transferasa (GST), capaces de inactivar las moleculas herbicidas a
través de su conjugación con glutation.
Por lo tanto, en este trabajo se ha evaluado el posible papel de las GSTs en la resistencia
de NTS a los inhibidores de ALS. Para ello se ha utilizado una población resistente a sulfonilureas
(inhibidores de ALS) que presenta resistencia NTS y una sensible de referencia. Se realizaron dos
experimentos, creciendo semillas de A. palmeri en placas verticales o en cultivo hidropónico
puro. Se ha evaluado la participación de las GST mediante la determinación de su acividad
enzimática, la biomasa, el contenido de tioles tras el tratamiento con diferentes dosis de una
sulfonilurea (nicosulfuron) aplicada sola o con un compuesto específico inhibidor de GST (NBDCl). Los resultados obtenidos sugieren cierta participación de las GSTs en la resistencia NTS, si
bien son experimentos preliminares que deben ser validados. [--]

Beraz, ALS inhibitzaileen aurrean NTS erresistentzian parte hartzen duten entzimek duten
erantzuna baloratzeko eta ikertzeko asmoa dago. Horretarako, bi populazio erabili dira, bata
sentikorra (NCS, Ipar Kalifornia jatorria) eta bestea NTS erresistentea (KSR, Kansas jatorria). Bi
esperimentu egin dira, A. palmeri haziak plaka bertikaletan eta ur-ontzietan (hidroponikoan).
GSTen parte-hartzea ebaluatu da, beren jarduera entzimatikoa, biomasa eta tiolen edukia
neurtuz, sulfonilurea (nicosulfuron) bakarrik edo GST inhibitzaile espezifiko (NBD-Cl) batekin
tratatu ondoren. Horrela, lortutako emaitzen bidez, GSTek KSR populazioko landareen NTS
erresistentzian nolabaiteko parte-hartzea dutela ikusi da, hala ere, baliozkotu beharreko atariko

weeds, and to avoid the evolution of other resistant biotypes, it is important to understand well
the resistance mechanism of these resistant weeds.
Amaranthus palmeri is a weed native to North America, and due to its physiology and
ability to spread and invade, it is considered a highly problematic weed and invasive plant. In
addition, numerous populations with resistance to different herbicides have been found, such as
resistance to herbicides that inhibit the enzyme acetolactate synthase (ALS). Generally, the
resistance mechanism is based on a less sensitive herbicide target, but there may also be
resistance mechanisms which are not related to the target (NTS), like the capacity of the plant to
metabolise the herbicide through glutathione-S-transferase (GST) enzymes, capable of inactivate
herbicide molecules through conjugation with glutathione.
Thus, in this work, the possible role of GSTs in the NTS resistance to ALS inhibitors has
been evaluated. For this propose, two populations have been used, one NTS resistant to
sulfonylureas (ALS inhibitors), and one sensitive. Two experiments were carried out: A. palmeri
seeds were sown in vertical plates and hydroponically. Participation of GSTs has been evaluated
through the analytical determination of their enzymatic activity, the biomass, and the thiol
content after the treatment with different doses of a sulfonylurea (nicosulfuron), alone or in
combination with a specific GST inhibitor (NBD-Cl). Obtained results suggest some participation
of GSTs in NTS resistance. These are preliminary experiments that should be validated, though. [--]
