This process is particularly relevant for the analysis of small and heterogeneous opulations, such as progenitor cells. In this study, the annotation of CD34+ cells is performed in samples from mobilized peripheral blood, obtained by Multiome 10X and CITE-Seq sequencing methods. The characterization of transcriptomic and membrane marker data enables the identification of different progenitor populations present in the samples. Additionally, different annotation methodologies are compared, including automatic annotation with SingleR and “Label Transfer”, as well as a manual annotation. Moreover, subpopulations within the hematopoietic stem cells (HSC) are identified, such as long-term HSC (LT-HSC) and short-term HSC (ST-HSC). Lastly, a Shiny application was used to facilitate the visualization of the annotated samples. Thus, this study provides new tools and insights to facilitate the analysis of hematopoietic stem and progenitor cells.
