La variedad Berués (Vitis vinifera L.), la cual se creía extinta, fue recuperada en la zona periurbana
de Pamplona en prospecciones recientes realizadas en esta zona. En este Trabajo Fin de Grado se
siguieron los principios FAIR y se utilizó la plataforma Galaxy para conocer el nivel de
variabilidad intravarietal existente en plantas de la variedad Berués a través de datos genómicos
obtenidos mediante Next Generation Sequencing.
En una primera fase del TFG, se usaron marcadores moleculares de tipo microsatélite en
combinación con bases de datos públicas para confirmar la identidad varietal de las plantas antes
de proceder a la secuenciación de su genoma. Posteriormente, y utilizando para ello datos
genotípicos de 18 mil SNPs en casi 800 variedades, se estudió el posicionamiento de la variedad
Berués en la estructura genética global de Vitis vinifera L. Se puso de manifiesto que dicha
variedad no pertenecía de manera inequívoca a una población geográfica concreta, aunque muy
posiblemente, esté relacionada con variedades de vinificación del Oeste de Europa.
La exploración genómica realizada mediante la plataforma de acceso abierto Galaxy, logró
identificar entre 10 y 14 millones de variantes entre las plantas secuenciadas y el genoma de
referencia de la vid. Más de la mitad de las variantes identificadas eran de tipo SNP, y el resto,
inserciones, deleciones y MNP (Multiple Nucleotide Polimorphisms). Además, se evaluaron los
efectos y el impacto que producían las variantes identificadas. La mayoría de variantes se
localizaron en regiones no codificantes (upstream, downstream y regiones intrónicas e
intergénicas) y no suponían un alto impacto en el genoma. En torno a un 0,1 % de las variantes
se clasificaron como de alto impacto, ya que modificaban secuencias codificantes y, por tanto,
podían afectar a la estructura de algunas proteínas. El análisis intravarietal resultó en la
identificación de 415.048 variantes comunes para las cinco muestras y multitud de variantes
únicas que podrían estar detrás de diferencias fenotípicas concretas. En conjunto, los resultados
obtenidos contribuirán a un mejor conocimiento de la variabilidad intravarietal existente en esta

The Berués variety (Vitis vinifera L.), which was thought to be extinct, was recovered in the

In the first phase of the TFG, microsatellite molecular markers were used in combination with
public databases to confirm the varietal identity of the plants before proceeding to the sequencing
of their genome. Afterwards, it was studied the positioning of the Berués variety in the global
genetic structure of Vitis vinifera L. using genotypic data of 18,000 SNPs in almost 800 varieties.
It became clear that this variety did not belong unequivocally to a specific geographic population,
although it is very possibly related to wine varieties from Western Europe.
The genomic exploration, carried out using the Galaxy open access platform, was able to identify
between 10 and 14 million variants between the sequenced plants and the reference genome of
the grapevine. More than half of the variants identified were SNPs, and the rest were insertions,
deletions and MNPs (Multiple Nucleotide Polymorphisms). In addition, the effects and impact of
the identified variants were evaluated. Most variants were located in non-coding regions
(upstream, downstream, intronic and intergenic regions) and did not have a high impact on the
genome. About 0.1 % of the variants were classified as high impact, as they modified coding
sequences and could therefore affect the structure of some proteins. The intravarietal analysis
resulted in the identification of 415,048 common variants for the five samples and a multitude of
unique variants that could be behind specific phenotypic differences. Overall, the results obtained
will contribute to a better understanding of the intravarietal variability in this variety. [--]
